| Abstract Cluster of differentiation 40 (CD40), the receptor for CD154, is a member of the tumor necrosis factor (TNF) receptor superfamily. Several studies have been conducted to investigate the effect of the CD40 rs1883832 polymorphism on atherosclerotic disease in different population; however, inconsistent results were obtained. In this study, we investigated the association of four polymorphisms (rs1883832, rs13040307, rs752118 and rs3765459) of CD40 gene and their effect on CD40 expression with the risk of ischemic stroke (IS) in a Chinese population. | Challenges to the gene concept have shown the difficulty of preserving the classical molecular concept, according to which a gene is a stretch of DNA encoding a functional product (polypeptide or RNA). The main difficulties are related to the overlaying of the Mendelian idea of the gene as a unit: the interpretation of genes as structural and/or functional units in the genome is challenged by evidence showing the complexity and diversity of genomic organization. | A Biologia Sintética, em franca expansão nos dias de hoje, consiste no uso de bioinformática e técnicas de engenharia genética e bioquímica com o objetivo de desenhar circuitos biológicos modulares, por meio do redirecionamento ou construção de novas rotas metabólicas e a criação de organismos artificiais, visando maximizar o seu funcionamento. Não se trata de uma técnica específica, mas de um conceito que pode utilizar diferentes abordagens de edição genômica para alcançar a meta final. Esse documento tem como objetivo fornecer uma visão abrangente desse novo ramo da biologia, de modo a ampliar a nossa percepção sobre as possibilidades futuras que podem ser vislumbradas sobre o desenvolvimento e as aplicações dessa nova abordagem científica. |

| This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Thus far, P. abyssi AP has been demonstrated to be the most thermostable AP, with half-lives at 100 and 105 degreesC of 18 and 5 h, respectively.... |

| Variações no número de cópias de regiões específicas do DNA, ou simplesmente CNVs (Copy Number Variations), constituem um tipo de marcador molecular que é encontrado com frequência nos genomas de mamíferos e plantas. Em humanos, o impacto das CNVs sobre os fenótipos já foi relatado em características adaptativas, mortalidade embrionária e em algumas doenças complexas como autismo, esquizofrenia, doença de Chron, artrite reumatoide, diabetes tipo I e obesidade. Such a goal has been reached in other biological systems through the development of molecular biology and cell culture. Indeed, development of a mollusc cell culture system is an essential step to approach many of the problems related to mollusc pathology, in particular to use molecular biology tools to study viral infection of these organisms.
